The National Institutes of Health (NIH) has changed its long-standing policy of accepting a given grant proposal only twice. The policy on the revision and timing of a resubmitted proposal remains the same; however, if a resubmitted proposal is not funded, that proposal can be submitted again as a new proposal without substantial changes in the scientific direction.

Of course, Research and Sponsored Programs (RSP) recommends contacting your program officer, talking to the RSP grant writer, and taking other steps to improve the competitiveness of your proposal, regardless of the submission cycle.
